What It Is
FOXO4-DRI is a modified peptide designed to selectively eliminate senescent cells, also called “zombie cells.” Senescent cells are damaged cells that stop dividing but refuse to die. They accumulate with age and release inflammatory signals that damage surrounding tissue and accelerate aging.
What It Does
FOXO4-DRI disrupts the interaction between FOXO4 and p53 proteins inside senescent cells, triggering apoptosis (programmed cell death) specifically in those cells while leaving healthy cells unharmed. In animal studies it reversed physical frailty, improved kidney function, restored fur density, and extended healthy lifespan.
How It Works
In senescent cells, FOXO4 binds to p53 and prevents it from triggering apoptosis. FOXO4-DRI is a modified peptide that competes with endogenous FOXO4 for p53 binding, disrupting this protective interaction and allowing p53 to initiate cell death in the senescent cell. Healthy cells are not affected because they do not rely on this FOXO4-p53 interaction for survival.
