What It Is
5-Amino-1MQ is a small molecule NNMT (Nicotinamide N-methyltransferase) inhibitor. NNMT is an enzyme that becomes overactive in fat tissue during obesity and aging, consuming NAD+ and promoting fat storage. Blocking NNMT restores NAD+ availability and shifts metabolism toward fat burning.
What It Does
5-Amino-1MQ reduces body fat, improves insulin sensitivity, increases NAD+ levels, activates SIRT1 (a longevity protein), and improves mitochondrial function. In animal studies it produced significant fat loss without changes to diet or exercise. It is particularly effective for visceral fat and metabolic dysfunction.
How It Works
By inhibiting NNMT, 5-Amino-1MQ prevents the enzyme from consuming NAD+. This raises intracellular NAD+ levels, which activates sirtuins and AMPK, shifting cellular metabolism toward fat oxidation and away from fat storage. It also reduces the size of fat cells and decreases fat cell proliferation.
Who Benefits Most
People with stubborn body fat, especially visceral fat. People with metabolic syndrome or insulin resistance. Anyone who wants to raise NAD+ levels through a different mechanism than direct supplementation. People interested in longevity and metabolic optimization.
Protocol and Dosing
Typical dose: 50 to 100 mg orally once daily. Can also be used as a sublingual or injectable compound. Cycle length: 4 to 12 weeks. Often stacked with NAD+ for synergistic effects.
| Route | Typical Dose | Frequency |
|---|---|---|
| Oral | 50 to 100 mg | Once daily |
Cautions
Limited long term human safety data. Use under practitioner supervision. Not recommended during pregnancy. Monitor liver function with extended use.
Pairs Well With
NAD+ for synergistic NAD+ restoration through complementary mechanisms. MOTS-C for comprehensive mitochondrial and metabolic optimization. Semaglutide or Tirzepatide for aggressive fat loss protocols.
